Metabolites are evaluated according to the EFSA PPR Guidance (2016) using the EFSA templates (2020) for assessing (Q)SAR reports and for summarizing and integrating the evidence. The forthcoming OECD Guidance on residue definition (2023) is expected to further demand additional RA/grouping. To assess relevance of impurities the Guidance on assessment of equivalence (2012) is followed.

Reflecting all recommendations, robust in silico predictions are generated for genotoxicity and other toxicity endpoints by complementary rule- and statistics-based (Q)SAR software tools. The reliability of the predictions is evaluated based on the prediction’s evidence. Knowledge of the chemistry/biology regarding the endpoint is needed for overall evaluation in a weight of evidence approach. The OECD QSAR Toolbox is additionally used for toxicity screening, assessment of organic functional groups, calculation of physico-chemical properties and similarity to facilitate grouping, selection of appropriate analogues and data gap filling.

Based on all information, a case is established with a rationale, and if needed with a testing proposal, to support the conclusion for dietary risk assessment of metabolites or for the evaluation of the relevance of impurities. State-of-the-art reports describe strategy, methods followed, and results obtained.

In summary, in silico methods are an efficient and accepted tool in the assessment of the toxicological relevance of pesticide metabolites and impurities.

• With the new release, a single sign-on function was introduced. The feature allows the use of Microsoft account to sign in into IUCLID with the same account. This feature does not require setting up separate user accounts in IUCLID.
• The new IUCLID version allows the use of PostgreSQL, an open source and free database which requires a lot of efforts for its initial set-up but less maintenance later.
• The public IUCLID API allows users to establish a connection between IUCLID and the organisation’s software. It can be used to develop new modules, plug-ins for UICLID, e.g. data analysis, data extractor, etc.
• IUCLID 6 enables document searches via a Universally Unique Identifier (UUID). Prior to the release, this function was possible for dossiers and entities only.
• With the release, new features for advanced import were introduced enabling administrators to modify the table of content for better representing of the dataset relevant for a subsequent import. Thus, only the records selected by the administrator and listed in the table of content can be later imported. The administrator can also see the recent modification date of the listed records per mouse rollover (hovering over the selected record).
• In the updated version, the user can see the number of attached files or entries in the modification history by a visual indicator in the respective study record.
• A new feature search by reference substance was introduced.
• The updated version enables a search in “identifiers”: When looking for a specific dossier with known UUID, the user should type the number in quotes to start a search (e.g. ”123”).
• The “Option of bulk delete” enables selecting and deleting of more than one record at once. In case the selected records are linked to another record, e.g. an endpoint summary, IUCLID shows a warning which is to be confirmed if the user wants to proceed with the deletion. In this case, IUCLID will delete both the record and the linkage.
• There is an improvement with regard to template linkage in datasets for e.g. read-across purposes: The new IUCLID version recognises the links set in source and target records. By deleting any template linked to the records in the datasets, IUCLID suggests removing all references and unlinking automatically. Previously, the user had to unlink templates or other references manually before deleting the record.
• The new version enables comparison of dossiers and mixtures. The user can choose dossiers for the same dataset and select the function generate report. This feature is especially helpful for biocidal users.
• The DNELs section has also undergone several modifications allowing several parameters to be displayed in the same row and introducing a shorter record less difficult to navigate through. Further changes are expected in the next IUCLID releases.
• ECHA plans a new service release in October 2022. The classic interface is to be completely removed with the upcoming release. Among the planned changes are improvements to the web interface, advanced settings for the report generator, support for BPR summary of product characteristics. A format update with the new OECD Harmonised Templates will follow in April 2023 and a new service release – in October 2023.
